ADE/PDE, OEL & Cleaning Validation

Toxicological risk assessment reports for shared manufacturing

Not every manufacturing question is answered by a standard PDE. When a cleaning agent residue, an intermediate, a degradation product or an unexpected carry-over needs to be assessed, we prepare a focused toxicological risk assessment that answers the specific question and documents the reasoning.

  • Typical questions Cleaning agents, intermediates, excursions, dedicated-facility decisions
  • Typical inputs Substance identity, amounts, exposure scenario
  • You receive Signed risk assessment report

Overview

About Toxicological Risk Assessment Reports

These assessments apply toxicology to manufacturing situations: setting limits for detergent residues, evaluating whether an excursion above a cleaning limit could harm patients, assessing degradation products formed during cleaning, or supporting a decision on whether a hazardous product needs dedicated facilities.

Each report states the question, the exposure scenario, the data used and the conclusion — with the uncertainties made explicit so that your quality unit can make and defend a decision.

Use cases

When this service helps

Cleaning agent residues

Health-based limits for detergents and their components.

Excursions above a cleaning limit

Patient-safety impact assessment to support batch disposition.

Degradation during cleaning

Evaluation of inactivated or degraded residues, for example from biologics.

Dedicated facility decisions

Toxicological input to decide whether shared manufacturing is acceptable.

Scope

What the service covers

  • Definition of the question and exposure scenario
  • Toxicological data review for the substance or its components
  • Limit derivation or comparison of actual exposure with a health-based limit
  • Assessment of uncertainty and any need for further data
  • Clear conclusion and recommendation for the quality decision

Who does what

The quality decision (for example batch release) remains with your quality unit. Globaxa provides the toxicological assessment that supports it.

Do cleaning agents need a PDE?

Cleaning agents are generally of lower toxicity than APIs, but their residues should still be justified. A health-based limit for the agent or its key components is a common and defensible approach.

Methodology

How we work

A defined sequence, agreed with you at the start, so you always know what happens next and what we need from you.

  1. Frame the questionWe agree exactly what needs to be answered and the exposure scenario.
  2. Assess the dataRelevant toxicology is reviewed for the substance or residue.
  3. ConcludeExposure is compared with a health-based limit and a conclusion is documented.

Deliverables

What you receive

Risk assessment report

Question, data, assessment and conclusion, signed by the toxicologist.

Summary for the quality record

A short statement suitable for deviation or change-control files.

Technical basis

Guidelines we work to

  • EMA HBEL guideline and Q&ACovers health-based limits, including for cleaning agents and intermediates.
  • EU GMP Chapters 3 and 5Require cross-contamination risks to be assessed using a toxicological evaluation.
  • ICH Q9(R1)Quality risk management principles that frame the decision.

What we need from you

  • Identity and composition of the substance or residue
  • Quantities involved and the products potentially affected
  • Dose and duration of the affected product
  • Relevant deviation or change-control records

Please share confidential information only after an NDA is in place — we are happy to sign one first.

Market and jurisdiction considerations

The approach follows EU/PIC/S and WHO expectations and is recognised internationally. Where a specific authority is involved (for example in an inspection response), we tailor the format to that authority.

FAQs

Toxicological Risk Assessment Reports: frequently asked questions

How quickly can an excursion be assessed?

Urgent excursion assessments are prioritised. The time needed depends on the data available for the substance; we confirm a realistic date when we receive the details.

How is this different from your impurity risk assessments?

These reports address manufacturing situations such as cleaning and cross-contamination. Impurity qualification in the drug substance or product is covered under Toxicological Risk Assessments.

Will the report support batch release?

It provides the toxicological basis for your quality unit’s decision; release decisions remain with your QP or quality head.
