ADE/PDE, OEL & Cleaning Validation
Occupational exposure band (OEB) assessment and categorisation
When a compound is new, or the data are thin, a numerical limit may not yet be possible. Occupational exposure banding (OEB) places the substance in a hazard category based on potency and toxicity, so you can choose appropriate handling and containment from the start.
- Typical use Early development, intermediates, new suppliers
- Typical inputs Structure, pharmacology, dose, any toxicology
- You receive OEB categorisation report with handling guidance

Overview
About OEB Assessment & Banding
Banding schemes typically use four to six bands, each corresponding to a range of airborne concentrations — from compounds with low hazard to highly potent substances that need closed handling. The exact band boundaries differ between companies and published schemes, so the scheme used should always be stated.
We assess pharmacological potency, toxicological hazards such as genotoxicity, reproductive toxicity and sensitisation, and any structural alerts. The band is documented with its rationale and the handling expectations that follow from it, and is revisited as more data are generated.
Use cases
When this service helps
Early development compounds
Set handling controls before toxicology data are complete.
Intermediates and impurities
Categorise substances that will never have a full data package.
Facility and equipment planning
Decide which suites need isolators or ventilated enclosures.
Supplier qualification
Understand the hazard of materials handled by contract partners.
Scope
What the service covers
- Review of potency (therapeutic dose, receptor activity) and available toxicity data
- Assessment of hazard flags: genotoxicity, carcinogenicity, reproductive toxicity, sensitisation
- Structural alerts and read-across from related compounds
- Band assignment under your company scheme or a stated published scheme
- Handling and containment expectations for the assigned band
- Trigger points for re-banding or deriving a numerical OEL
Who does what
Banding is a desk-based hazard assessment. It supports, but does not replace, site-specific risk assessments carried out by your EHS function.
Is OEB 5 the same everywhere?
No. Band numbers and concentration ranges vary between schemes. A compound described as “OEB 5” by one company may fall into a different band elsewhere, so reports always state the scheme and the concentration range.
Methodology
How we work
A defined sequence, agreed with you at the start, so you always know what happens next and what we need from you.
- Collect informationStructure, intended dose, pharmacology and any available data.
- Assess hazardsPotency and key toxicological endpoints are evaluated.
- Assign the bandThe compound is placed in a band with a written rationale.
- Define handlingContainment and procedural expectations are listed for the band.
Deliverables
What you receive
OEB report
Band assignment, rationale and data gaps for each compound.
Handling guidance
Practical containment, PPE and procedural expectations for the band.
Portfolio summary
A table of all banded compounds for site-wide planning.
Technical basis
Guidelines we work to
- ISPE Risk-MaPPDescribes risk-based approaches to exposure control in pharmaceutical manufacturing.
- Published banding schemesSeveral industry and occupational-health bodies publish banding approaches; band ranges must be stated because they are not harmonised.
What we need from you
- Chemical structure and identity
- Intended or known therapeutic dose and mechanism of action
- Any toxicology, pharmacology or safety data
- Your company banding scheme, if you have one
Please share confidential information only after an NDA is in place — we are happy to sign one first.
Market and jurisdiction considerations
Banding is primarily a worker-safety and facility-design tool rather than a regulatory submission. It is, however, frequently reviewed during customer audits and GMP inspections of potent-compound facilities.
FAQs
OEB Assessment & Banding: frequently asked questions
Can a band be converted into a numerical OEL later?
Yes. As toxicology and clinical data accumulate, the band can be refined into a numerical OEL using the same evaluation.
Do you use our company’s banding scheme?
Yes, if you have one. Otherwise we use a published scheme and state its ranges clearly.
Does banding affect cleaning validation?
Banding concerns worker exposure. Cleaning limits are based on a PDE/ADE, although highly potent compounds often need both.
